Biomarker: G005-IgE Rye Grass, Perennial

Overview

G005-IgE measures immunoglobulin E (IgE) antibodies in blood that specifically recognize pollen from perennial rye grass (Lolium perenne). IgE is an antibody involved in immediate-type allergic reactions, including allergic rhinitis and asthma symptoms after inhalation of sensitizing pollen. A detectable or elevated result indicates sensitization to perennial rye grass but does not by itself confirm symptomatic allergy or predict reaction severity. The test is clinically useful when evaluating seasonal respiratory symptoms, identifying relevant pollen sensitization, and supporting decisions about allergen avoidance or immunotherapy in conjunction with the clinical history.

Clinical Use Cases

  • Evaluation of suspected seasonal allergic rhinitis or conjunctivitis associated with grass-pollen exposure.
  • Assessment of possible grass-pollen-triggered asthma or other allergic respiratory symptoms.
  • Identification of perennial rye grass sensitization when skin testing is unsuitable, contraindicated, or affected by medications or skin disease.
  • Selection of allergens for environmental avoidance counseling or allergen immunotherapy.
  • Research on grass-pollen sensitization, including relationships between rye grass pollen exposure and seasonal or thunderstorm-related asthma.

Specimen Types

  • Venous blood, with allergen-specific IgE measured in serum.
  • Plasma may be accepted by some validated laboratory assays, depending on the assay manufacturer and laboratory protocol.

Measurement Methods

  • In-vitro allergen-specific IgE immunoassay performed on serum or plasma.
  • Fluorescent enzyme immunoassay.
  • Enzyme-linked immunosorbent assay (ELISA).
  • Chemiluminescent immunoassay.
  • Radioallergosorbent testing (RAST), an older method that is now largely replaced by newer immunoassays.
  • Component-resolved testing for individual perennial rye grass allergens, such as Lol p 1 or Lol p 5, may be used in specialized clinical or research settings.

Test Preparation and Influencing Factors

  • No fasting or other special preparation is generally required for an allergy blood test.
  • Patients should provide a complete list of prescription medicines, over-the-counter medicines, vitamins, and supplements.
  • Antihistamines generally do not interfere with serum-specific IgE testing, although they can suppress responses during skin testing.
  • Medications should not be stopped unless directed by a healthcare professional.
  • Recent or ongoing exposure to rye grass pollen may influence symptom correlation, but a positive specific IgE result indicates sensitization rather than proving that exposure causes symptoms.
  • Total IgE concentration, age, immune status, assay characteristics, and the laboratory’s reporting scale can affect interpretation.
  • Cross-reactivity among grass pollens can produce reactivity to multiple grass extracts, so results should be interpreted with geographic exposure, seasonality, symptoms, and other allergy testing.
  • The concentration of perennial rye grass-specific IgE does not reliably predict the severity of a future allergic reaction.
